Topographic Genotyping

Topographic Genotyping

US Patent:
6340563, Jan 22, 2002
Filed:
Jun 24, 1996
Appl. No.:
08/667493
Inventors:
Sydney David Finkelstein - Aspinwall PA 15215
Patricia Anne Finkelstein - Aspinwall PA 15215
International Classification:
C12Q 168
US Classification:
435 6
Abstract:
A method for topographic genotyping is described. The method comprises the steps of placing a biological specimen having DNA of a patient under a microscope. Then there is the step of inspecting the biological specimen microscopically with the microscope. Next there is the step of choosing a microscope size target on the biological specimen based on its histopathologic characteristics. Next there is the step of separating the target from the specimen. Then there is the step of obtaining DNA sequences from the target so the DNA sequences can be amplified. Next there is the step of amplifying the DNA sequences. Then there is the step of detecting mutations in the DNA sequences. More specifically, the method comprises the steps of separating a section from a specimen of fixative treated tissue. Then there is the step of obtaining DNA sequences from the section. Next there is the step of amplifying the DNA sequences by cycling them in a PCR machine, with each cycle heating them to a temperature no greater than 99° C.

Molecular Analysis Of Cellular Fluid And Liquid Cytology Specimens For Clinical Diagnosis, Characterization, And Integration With Microscopic Pathology Evaluation

US Patent:
2006014, Jun 29, 2006
Filed:
Oct 24, 2005
Appl. No.:
11/256150
Inventors:
Sydney Finkelstein - Pittsburgh PA, US
Patricia Swalsky - Pittsburgh PA, US
International Classification:
C12Q 1/68
US Classification:
435006000
Abstract:
The application relates to methods, materials, kits, and devices for characterizing fluid specimens of any type or source that may or may not contain cells. The methods, materials, kits, and devices can be used for analyzing fluid obtained from various organs or near a purported tumor site, such as via breast ductal lavage or aspiration of a pancreatic cyst. The methods, materials, kits, and devices generate molecular information that can be used in conjunction with clinical, imaging, and microscopic pathology evaluation to provide a superior pathology diagnosis for clinical and research.

The method comprises the steps of separating a section from a specimen of fixative treated tissue. Then there is the step of obtaining DNA sequences from the section. Next there is the step of amplifying the DNA sequences by cyphaling them in a PCR machine, with each cycle heating them to a temperature no greater than 99° C., and then back to a temperature of 55° C. in 5 minutes. Next there is the step of detecting mutations in the DNA sequences. Preferably, the separating step includes the step of cutting one to three 2-6 micron thick histeologic sections from the specimen.
