Methods For Inducing Mixed Donor-Recipient Chimerism In An Allograft Transplant Recipient

US Patent:
2010025, Oct 7, 2010
Filed:
Feb 24, 2010
Appl. No.:
12/711836
Inventors:
Maria Z. Siemionow - Cleveland OH, US
Assignee:
The Cleveland Clinic Foundation - Cleveland OH
International Classification:
A61K 39/395, A61P 37/06
US Classification:
4241731
Abstract:
A method is provided for obtaining mixed donor-recipient chimeric cells, comprising the steps of (a) administering to an allograft recipient a short-term immunosuppressive regimen that depletes about 50% to about 99.9% of recipient T cells circulating in the peripheral blood of the recipient; (b) implanting an allograft from a donor into the recipient; (c) allowing the development of mixed donor-recipient chimeric cells in the recipient after completion of the immunosuppressive regimen; and (d) harvesting the chimeric cells from the recipient. The invention further provides method for compiling a library of chimeric cells and using the library to select of chimeric cells for the induction of tolerance to an allograft in a naïve or previously transplanted recipient.

Use Of Epineural Sheath Grafts For Neural Regeneration And Protection

US Patent:
2011008, Apr 14, 2011
Filed:
Apr 2, 2009
Appl. No.:
12/935232
Inventors:
Maria Siemionow - Shaker Heights OH, US
Krzysztof Siemionow - Shaker Heights OH, US
International Classification:
A61F 2/02, A61B 17/32
US Classification:
623 2372, 623 1111, 606170
Abstract:
Described herein is conduit material that causes minimal inflammatory reaction, and serves as a structural guide for regenerating nerve tissue (e.g., axons). Thus, the invention is directed to methods of treating a nerve injury in an individual in need thereof. The methods employ an isolated, naturally occurring epineural sheath, and can be used, for example, to regenerate nerve tissue in an individual in need thereof. Also provided herein is a device for harvesting an epineural sheath.

Methods Of Engineering Neural Tissue

US Patent:
2012017, Jul 5, 2012
Filed:
Sep 1, 2010
Appl. No.:
13/393750
Inventors:
Maria Siemionow - Shaker Heights OH, US
International Classification:
A61K 35/30, C12N 5/079
US Classification:
424 937, 435325
Abstract:
In one aspect, the invention is a method of generating a neural conduit comprising neurotrophic factors and angiogenic factors ex vivo comprising introducing cells that enhance nerve regeneration into an isolated, naturally occurring epineural sheath, thereby producing a combination. The combination is maintained under conditions in which neurotrophic factors and angiogenic factors are expressed in the epineural sheath, thereby generating a neural conduit comprising neurotrophic factors and angiogenic factors ex vivo. In another aspect, the invention is directed to neural conduits such as a neural conduit produced by the methods provided herein. In other aspects, the invention is directed to an article of manufacture.

Induction And Maintenance Of Tolerance To Composite Tissue Allografts

US Patent:
2004000, Jan 8, 2004
Filed:
Apr 30, 2003
Appl. No.:
10/427013
Inventors:
Maria Siemionow - Cleveland OH, US
International Classification:
A61K039/395
US Classification:
424/141100, 424/144100
Abstract:
The present invention provides a clinically-applicable approach for inducing long-term, donor-specific tolerance to donor antigens, especially in recipients of CTA and/or solid organ transplants, without the requirement for patient preconditioning, without the need for chronic immunosuppressive regimens, and without the occurrence of GVHD. In particular, a method is provided for inducing donor-specific tolerance in a semi-allogeneic or a fully-allogeneic transplant recipient by administering to the recipient a therapeutically effective amount of an immunosuppressive agent that depletes T cells and a therapeutically effective amount of anti- T cell receptor antibodies, and implanting an allograft into the recipient.
