Combination Bacteriolytic Therapy For The Treatment Of Tumors

US Patent:
7344710, Mar 18, 2008
Filed:
Nov 21, 2002
Appl. No.:
10/495116
Inventors:
Long Dang - Ann Arbor MI, US
Kenneth W. Kinzler - Bel Air MD, US
Bert Vogelstein - Baltimore MD, US
Assignee:
The Johns Hopkins University - Baltimore MD
International Classification:
A01N 63/00, A61K 48/00
US Classification:
424 931, 424 932, 424 9341
Abstract:
Current chemotherapeutic approaches for cancer are in part limited by the inability of drugs to destroy neoplastic cells within poorly vascularized compartments of tumors. We have here systematically assessed anaerobic bacteria for their capacity to grow expansively within avascutar compartments of transplanted tumors. Among 26 different strains tested, one () appeared particularly promising. We created a strain of devoid of its lethal toxin (-NT) and showed that intravenously injected -NT spores germinated within the avascular regions of tumors in mice and destroyed surrounding viable tumor cells. When -NT spores were administered together with conventional chemotherapeutic drugs, extensive hemorrhagic necrosis of tumors often developed within 24 hours, resulting in significant and prolonged anti-tumor effects. This strategy, called combination bacteriolytic therapy (COBALT), has the potential to add a valuablle dimension to the treatment of cancer.

Assignee:
THE REGENTS OF THE UNIVERSITY OF MICHIGAN - Ann Arbor MI
NEVADA CANCER INSTITUTE - Las Vegas NV
International Classification:
C12Q 1/68, C12Q 1/37
US Classification:
435 6, 435 24
Abstract:
The present invention provides methods and systems for the detection of tumor vessel response to antiangiogenic therapies. The present invention also provides compositions and methods for therapeutic and research applications. In particular, the present invention provides systems and methods that employ CD26, HIF-1, and HIF-1 pathway components as biomarkers for monitoring antiangiogenic therapies and as therapeutic targets.
