Method For Determining The Immunocompetence Of A Mammal And Therapies Related Thereto

US Patent:
6197531, Mar 6, 2001
Filed:
Jan 21, 1998
Appl. No.:
9/009958
Inventors:
Judy Lieberman - Brookline MA
Assignee:
Center For Blood Research, Inc. - Boston MA
International Classification:
G01N 3353
US Classification:
435 724
Abstract:
A method for determining the immunocompetence of a mammal is described. Bodily fluid having T cell receptor complexes from a mammal is provided. Expression of a first component and a second component of the T cell receptor complexes is evaluated. An abnormal ratio of the number of cells expressing the first component as compared to the number of cells expressing the second component indicates altered immunocompetence in the mammal. Also described are methods for monitoring a therapeutic treatment of a disease in a mammal associated with an abnormal level of a T cell receptor complex component, methods for assessing the progression of a disease in a mammal associated with an abnormal level of a T cell receptor complex component, methods for predicting the prognosis of a mammal having a disease associated with an abnormal level of a T cell receptor complex component, methods for determining if a mammal is at risk for AIDS, methods for evaluating an agent for use in treating HIV infection, and methods for treating HIV infection in a mammal. Pharmaceutical compositions and kits are also provided.

Abstract:
The present invention is based, at least in part, on the discovery of compositions and methods for the treatment and prevention of infectious diseases or disorders, e. g. , HIV infection, AIDS, and AIDS-related diseases. In particular, the present invention pertains to methods of modulating cellular gene expression or protein activity, e. g. , CCR5, gene expression or protein activity and/or gene expression or protein activity of a gene or sequence of an infectious agent, in order to treat or prevent infectious diseases or disorders, HIV infection, AIDS, or an AIDS-related disease or disorder. In one embodiment the combination of an RNA interfering agent targeting a cellular gene in combination with an RNA interfering agent targeting a gene or sequence of an infectious agent results in prolonged prevention of infection by an infectious agent. The present invention is based on the identification of novel RNA interference agents, e. g. , siRNA molecules, which target cellular genes, e. g.

Sirna Microbicides For Preventing And Treating Diseases

US Patent:
7943589, May 17, 2011
Filed:
Jun 5, 2006
Appl. No.:
11/916334
Inventors:
Judy Lieberman - Brookline MA, US
Deborah Palliser - Cambridge MA, US
David Knipe - Auburndale MA, US
Assignee:
President and Fellows of Harvard College - Cambridge MA
Immune Disease Institute, Inc. - Boston MA
International Classification:
A61K 31/70, C07H 21/04, C07H 21/02, C12Q 1/68
US Classification:
514 44A, 536 231, 536 241, 536 245, 435 6, 435375
Abstract:
The invention provides a microbicidal composition comprising at least one siRNA. The siRNA is an RNA duplex made of one or two molecules. A portion of the siRNA is identical to a target sequence in an essential gene of a virus. The virus may be a herpesvirus, for example, HSV-1 or HSV-2. Preferably, the herpesvirus is HSV-2. The microbicidal composition further comprises a pharmaceutically acceptable carrier. Also included in the invention are methods to prevent and treat viral infections by administration of the microbicidal composition. Preferably, the microbicidal composition is administered transmucosally.

Therapeutic And Diagnostic Strategies

US Patent:
2012005, Mar 8, 2012
Filed:
Sep 18, 2009
Appl. No.:
13/063155
Inventors:
Judy Lieberman - Brookline MA, US
Ashish Lal - Brookline MA, US
Dipanjan Chowdhury - Chestnut Hill MA, US
Assignee:
IMMUNE DISEASE INSTITUTE, INC. - Boston MA
International Classification:
A61K 31/7105, A61P 37/00, A61P 37/06, C12N 5/071, A61P 35/00
US Classification:
514 44 A, 435375
Abstract:
The present invention encompasses the finding that microRNAs (miRNAs) regulate certain key proteins involved in DNA repair. In some embodiments, a miRNA suppresses levels and/or activity of one or more DNA repair proteins. In some such embodiments, such suppression renders cells hypersensitive to certain DNA damage agents (e.g., γ-irradiation and genotoxic drugs, among others). The present invention provides various reagents and methods associated with these findings including, among other things, strategies for treating cell proliferative disorders, certain diagnostic systems, etc.
