Apparatus And Method For Sterilizing, Seeding, Culturing, Storing, Shipping, And Testing Tissue, Synthetic, Or Mechanical Heart Valves Orvalve Segments

US Patent:
5846828, Dec 8, 1998
Filed:
Jun 7, 1995
Appl. No.:
8/478309
Inventors:
Alvin Peterson - Jamul CA
Lee K. Landeen - San Diego CA
John Bennett - San Diego CA
Jason Gee - San Diego CA
Scott Chesla - Atlanta GA
Joan Zeltinger - San Diego CA
Assignee:
Advanced Tissue Sciences - La Jolla CA
International Classification:
C12N 500
US Classification:
435399
Abstract:
An apparatus and method for sterilizing, seeding, culturing, storing, shipping, and testing heart valves is disclosed. Specifically, the present invention relates to an apparatus and method for dynamically seeding and culturing heart valves with human cells, resulting in a heart valve populated with viable human cells. In a preferred embodiment, the apparatus includes a fluid reservoir, a pneumatic pressure chamber, a pressure source for providing a varying pressure to the pressure chamber, and a heart valve holder. In an alternative exemplary embodiment, the apparatus includes a fluid reservoir, a pump, a venturi tube, a heart valve chamber, a heart valve holder, and a valve connected to a timer. In each embodiment, the components are combined in a fluid circuit to create a variable cycling and pulsatile fluid flow for seeding and culturing which can closely resemble the physiological conditions found in the human heart.

Assignee:
Advanced Tissue Sciences, Inc. - La Jolla CA
International Classification:
C12M 300, C12N 500
US Classification:
4352841
Abstract:
An apparatus and method for sterilizing, seeding, culturing, storing, shipping, and testing three-dimensional tissue constructs is disclosed. The apparatus includes a fluid reservoir, a pump, at least one treatment chamber, and a means for controlling media flow characteristics around a tissue construct disposed within the treatment chamber, and for controlling movement of the construct itself, so as to simulate a variety of physiologic conditions. One exemplary embodiment of the invention includes a means for applying an axial stress to the construct. Applying an axial stress to the construct during seeding and culturing results in a tissue-engineered construct with cells and their fibers oriented in a manner which is more likely to possess long term dimensional stability and the patency of native vessels with normal physiologic function.

Apparatus For The Growth And Packaging Of Three Dimensional Tissue Cultures

US Patent:

Filed:
Feb 5, 1996
Appl. No.:
8/597053
Inventors:
Dawn Orton Applegate - San Diego CA

Assignee:
Advanced Tissue Sciences, Inc. - La Jolla CA
International Classification:
C12M 300
US Classification:
4352841
Abstract:
A tissue culture chamber is disclosed. The chamber is a casing that provides for growth of three-dimensional tissue, and more specifically for the growth of skin tissue, that can be grown, preserved in frozen form, and shipped to the end user in the same aseptic container. The tissue culture chamber includes a casing comprising a substrate within the casing designed to facilitate three-dimensional tissue growth on the surface of the substrate. The casing includes an inlet and an outlet port which assist the inflow and outflow of media. The casing also includes at least one flow distributor. In one embodiment, the flow distributor is a baffle, which is used to distribute the flow of the media within the chamber to create a continuous, uniform piece of three-dimensional tissue. In a second embodiment, the flow distributor is a combination of deflector plates, distribution channels, and a flow channel. In each embodiment, the casing further includes a seal so as to ensure an aseptic environment inside the chamber during tissue growth and storage.
