Methods And Compositions For Modulation Of T-Cells Via The Kynurenine Pathway

US Patent:
2012025, Oct 4, 2012
Filed:
Nov 19, 2010
Appl. No.:
13/510655
Inventors:
Joel D. Ernst - New York NY, US
Ludovic Desvignes - New York NY, US
International Classification:
A61K 31/196, C12Q 1/02, A61P 37/06, A61K 31/198, A61P 29/00, C12N 5/07, G01N 33/53
US Classification:
514564, 435375, 435 29, 435 792, 514567
Abstract:
The present invention provides methods for the modulation of T cells and T cell responses, particularly of Th17 effector cells. The invention provides methods of and compositions for modulating T cells, particularly T cells expressing IL-17, particularly Th17 effector cells, via the tryptophan metabolism pathway, particularly using tryptophan metabolites, kynurenines and kynurenine analogs or metabolites. The invention provides assays for screening Th17 modulators and kynurenine analogs or compounds.

Epitope Therapy For Infectious Diseases

Epitope Therapy For Infectious Diseases

US Patent:
2013024, Sep 19, 2013
Filed:
Aug 18, 2011
Appl. No.:
13/811915
Inventors:
Joel D. Ernst - New York NY, US
Tyler D. Bold - Boston MA, US
Assignee:
NEW YORK UNIVERSITY - New York NY
International Classification:
A61K 39/04
US Classification:
4241851, 4241841, 42419711, 4242481, 530326
Abstract:
The present invention relates to a method of treating a subject for an infectious disease. This method involves selecting a subject infected with an infectious disease-causing agent. The infectious disease-causing agent elicits, in the selected subject, an immune response that is insufficient to cure the selected subject of the infectious disease. The method also involves administering to the selected subject a synthetic peptide representing an antigen epitope associated with an immune response to the infectious agent. The immune response to the synthetic peptide is sufficient to treat the selected subject for the infectious disease.

US Patent:
7566459, Jul 28, 2009
Filed:
Feb 27, 2006
Appl. No.:
11/362539
Inventors:
Joel D. Ernst - New York NY, US
Niaz Banaiee - New York NY, US
Assignee:
New York University - New York NY
International Classification:
A61K 39/04, A61K 49/00, C07H 21/04, C07H 21/02
US Classification:
4242481, 536 231, 536 237, 424 91, 424 92, 424 931, 424 932, 4242341, 4242781
Abstract:
The present invention features modified strains which lack a functional lspA gene, and vaccine formulations comprising the same. In many embodiments, a modified strain of the invention comprises deletion or inactivation of the lspA gene and/or one or more non-lspA genes that are involved in inhibition of macrophage responses to IFNγ. The present invention also features methods of using the modified strains to treat or present infection in mammals. In addition, the present invention features methods of using ΔlspA strains to identify non-lspA genes or other cellular components that are involved in proinflammatory stimulation of macrophages or inhibition of macrophage responses to IFNγ.
