Molecular Biology: DNA/RNA isolation, purification and amplification. PCR cloning with TA/Gateway technology. Co-immunoprecipitation and GST pull down assay. Quantitative PCR with SYBR Green technique. Construct design and cloning to achieve overexpression and knockdown (shRNA) of targeted genes in cell lines and animals via adenovirus and lentivirus. Western blotting, In situ Western blotting, ELISA, Flow cytometry, in vitro transcription, protein engineering, exosome isolation and characterization Cellular Biology: Culture of cancer cell lines (HepG2, Hep3B, MCF7, HEK cells). Establishment of stable transfected cell line. In vitro cancer assay: proliferation assay (Staining with proliferation markers: Ki67, Phospho-Histone 3; Flow cytometry with DAPI staining for cell cycle analysis; High-through put screening with CellTiter Glo luminescent assay); cell death assay (Caspase 3 immunoblotting, Flow cytometry assay with Annexin 5 staining, MTT assay); cell function assay: Mitochondrial function assay and lysotracker function. Cellular and Animal Imaging: Harvesting, fixing, trimming, processing, embedding, sectioning of animal and human samples. Immunohistochemistry staining. Immunofluorescence staining, Laser scanning confocal microscopy (running the confocal imaging core in CMRL), living cell imaging, organelle imaging; 2D echocardiography and color Doppler in cardiac imaging and function assessment. Whole animal imaging and non-invasive tracking of implanted cells. In Vivo: Collection of serum/plasma/whole blood, tissues, primary tumor material and metastases from animal models. Mouse anesthesia and surgery. Tumor xenotransplantation in immunosuppressed nude mice, Pharmacological agent administration to mice or rats via IV, IP, SC, IM and PO. Establishment of plasma cell tumor transgenic mouse model. Software: GraphPad Prism, SPSS.
