Single cell cloning, bulk cloning (by serial dilution, cloning rings, or sorting) Immunostaining/Imaging Fluorescent Immunocytochemistry (preparation of fixatives, detergent solutions, blocking agents; fluorescent multicolor labeling, up to 4 simultaneous fluorophores; ReAsh labeling for tetracysteine-tagged DNA; biotin/streptavidin and HRP expression amplification; labeling with Quantum Dots). Confocal microscopy and live-imaging of eukaryotic cell samples (skilled with Olympus Fluoview1000/Zeiss LSM 510 microscopes) Image processing (highly skilled in Adobe Photoshop, ImageJ, Bitplane Imaris, Zeiss LSM software) Immunohistochemistry on paraffin embedded blocks (DAB staining) Preparation of samples for electron microscopy (photooxidation of samples using FRET with ReAsh/DAB method, staining and counter staining with osmium tetroxide and uranyl acetate, embedding with Durcupan resin) Molecular/Biochemical/Misc Western Blots (PAGE) (semi dry transferring, probing with HRP secondaries, ECL detection and GraphPad Prism data analysis) Transformation (DH5alpha and XL1Blue), inoculation, small, medium and large scale culture for plasmid DNA isolation (Quiagen kits). Sub-cloning (mcherry vector - PCR, sequencing etc) Surgical isolation of hepatocytes from mice (post mortem) Basic script writing (MatLab software) Graduate Teaching Assistant/Demonstrator (University College London) Home Office Personal License (UK Animal Handling certificate)
