Disclosed are compositions and methods for quantitation and calibration of images in fluorescence microscopy. Also provided are tissue phantoms that contain known amount(s) of fluorophore standard(s), as well as components and diagnostic kits containing the same for use in various histological analyses. In certain embodiments, three distinct nucleic-acid based assays provide improvements over conventional TUNEL methods to facilitate precise quantitation of a variety of nucleic acids obtained from a biological sample.

Several types of DNA cuts are used as markers of apoptosis for detection of apoptotic cells in situ. The present invention includes a method to detect DNase II type DNA damage, bearing 5′OH using vaccinia topoisomerase I. The present invention also includes a method combining a ligase-based method to detect DNase I type DNA damage and the topoisomerase based method, resulting in simultaneous detection of two specific types of DNA damage in situ.

Neurologic dysfunction resulting from stroke is diminished by administering to a patient an effective amount of a tetra-substituted pyrazine over a relatively continuous regimen. Conveniently, an initial bolus is administered to bring the level of the drug up to a pre-determined blood level, followed by continuous infusion employing a pump or by continuous intravenous infusion.
