Intron-Based Assay For Detecting And Characterizing Chromosomal Rearrangement

US Patent:
6183969, Feb 6, 2001
Filed:
Apr 15, 1999
Appl. No.:
9/293569
Inventors:
Abram Gabriel - Princeton NJ
Assignee:
Rutgers, The State University of New Jersey - New Brunswick NJ
International Classification:
C12Q 168
US Classification:
435 6
Abstract:
An assay is provided for detecting and characterizing chromosomal rearrangements in eukaryotic cells by genetically engineering a eukaryotic cell to contain a selectable/counter-selectable marker gene with a site for generating a double strand break within an intron of the eukaryotic cell; generating the double strand break; detecting cells which repair the double strand break by chromosomal rearrangements; isolating DNA from the detected cells; and characterizing the chromosomal rearrangement as a nonhomologous translocation, inversion, deletion or insertion.

Method For Sequencing Nucleic Acid Sequences Without Chain Termination

US Patent:
7264931, Sep 4, 2007
Filed:
Sep 5, 2001
Appl. No.:
10/344891
Inventors:
Abram Gabriel - Princeton NJ, US
Yoshifumi Kobayashi - Highland Park NJ, US
Assignee:
Rutgers, the State University of New Jersey - New Brunswick NJ
International Classification:
C12Q 1/68, C12P 19/34, C07H 19/00
US Classification:
435 6, 435 912, 536 221
Abstract:
A method is provided for determining the sequence of a nucleic acid sequence via incubation of a primer extended product of the nucleic acid sequence in separate pools of semi-random oligonucleotides of 5′ (N)A3′, 5′ (N)G3′, and 5′ (N)T3′, wherein N is A, C, G or T and n is selected so that the oligonucleotides will optimally anneal to all complementary sequences.
